Remove the hose/tube at the inlet of the carb.
...Hold your hand against the carb inlet while cranking the engine.
...Do you feel a strong suction?

After hand choking, are the spark plugs wet with fuel?
...If not, there may be a vacuum leak in the intake?

Water in the gas? ...Fresh gas, not old gas that has lost most of its volatility?

The low compression in one cylinder should not prevent the engine from starting.
