Transmission in neutral, both tires will most likely turn in the same direction. The motion will take the path of least resistance, and may only turn the transmission output shaft. Put transmission in gear. Turning one tire can't turn the pinion gear so the movement must transfer thru the differential spider gears and turn the opposite tire backwards.
If putting the transmission in gear makes it so you cannot turn the tire, something in the differential has come loose or gotten jammed in there and locked up the spider gears.
